Spatio-Temporal Modelling of Perfusion Cardiovascular MRI
Myocardial perfusion MRI provides valuable insight into how coronary artery and microvascular diseases affect myocardial tissue. Stenosis in a coronary vessel leads to reduced maximum blood flow (MBF), but collaterals may secure the blood supply of the myocardium but with altered tracer kinetics. To date, quantitative analysis of myocardial perfusion MRI has only been performed on a local level, largely ignoring the contextual information inherent in different myocardial segments. This paper proposes to quantify the spatial dependencies between the local kinetics via a Hierarchical Bayesian Model (HBM). In the proposed framework, all local systems are modelled simultaneously along with their dependencies, thus allowing more robust context-driven estimation of local kinetics. Detailed validation on both simulated and patient data is provided
Bayesian Age-Period-Cohort Modeling and Prediction - BAMP
The software package BAMP provides a method of analyzing incidence or mortality data on the Lexis diagram, using a Bayesian version of an age-period-cohort model. A hierarchical model is assumed with a binomial model in the first-stage. As smoothing priors for the age, period and cohort parameters random walks of first and second order, with and without an additional unstructured component are available. Unstructured heterogeneity can also be included in the model. In order to evaluate the model fit, posterior deviance, DIC and predictive deviances are computed. By projecting the random walk prior into the future, future death rates can be predicted.
Spatially regularized estimation for the analysis of DCE-MRI data
Competing compartment models of different complexities have been used for the quantitative analysis of Dynamic Contrast-Enhanced Magnetic Resonance Imaging data.
We present a spatial Elastic Net approach that allows to estimate the number of compartments for each voxel such that the model complexity is not fixed a priori.
A multi-compartment approach is considered, which is translated into a
restricted least square model selection problem. This is done by
using a set of basis functions for a given set of candidate rate
constants. The form of the basis functions is derived from a kinetic
model and thus describes the contribution of a specific compartment.
Using a spatial Elastic Net estimator, we chose a sparse set of basis functions per voxel, and hence, rate constants of compartments.
The spatial penalty takes into account the voxel structure of an image and performs better than a penalty treating voxels independently.
The proposed estimation method is evaluated for simulated images and applied to an in-vivo data set

We present a nonnegative Elastic Net approach for the analysis of Dynamic Contrast-Enhanced Magnetic Resonance Imaging data. A multi-compartment approach is considered, which is translated into a (restricted) least square model

Using the Elastic Net estimator, we chose clusters of basis functions, and hence, rate constants of compartments. As further challenge, the estimator has to be restricted to positive regression parameters, which correspond to transfer rates of the compartments. Bayesian modelling of space-time interactions on the Lexis diagram
We propose a full model-based framework for a statistical analysis of incidence or mortality count data stratified by age, period and space, with specific inclusion of additional cohort effects. The setup will be fully Bayesian based on a series of Gaussian Markov random field priors for each of the components. Additional space-time interactions will be either modelled as space-period or space-cohort effects. Statistical inference is based on efficient algorithms to block update Gaussian Markov random fields, which have recently been proposed in the literature. We illustrate our approach in an analysis of stomach cancer data in West Germany
A Semi-parametric Technique for the Quantitative Analysis of Dynamic Contrast-enhanced MR Images Based on Bayesian P-splines
Dynamic Contrast-enhanced Magnetic Resonance Imaging (DCE-MRI) is an
important tool for detecting subtle kinetic changes in cancerous tissue.
Quantitative analysis of DCE-MRI typically involves the convolution of an
arterial input function (AIF) with a nonlinear pharmacokinetic model of the
contrast agent concentration. Parameters of the kinetic model are biologically
meaningful, but the optimization of the non-linear model has significant
computational issues. In practice, convergence of the optimization algorithm is
not guaranteed and the accuracy of the model fitting may be compromised. To
overcome this problems, this paper proposes a semi-parametric penalized spline
smoothing approach, with which the AIF is convolved with a set of B-splines to
produce a design matrix using locally adaptive smoothing parameters based on
Bayesian penalized spline models (P-splines). It has been shown that kinetic
parameter estimation can be obtained from the resulting deconvolved response
function, which also includes the onset of contrast enhancement. Detailed
validation of the method, both with simulated and in vivo data, is provided
